为了解竹子对大气O3浓度升高的生理响应规律及耐受O3胁迫能力的种间差异性,为气候变化背景下的竹林培育应对策略提供理论依据,运用开顶式同化箱(OTCs)开展了5个O3浓度梯度(过滤大气CF、环境大气NF、50、100和150 nl·L-1)对毛竹(Phyllostachys edulis)和四季竹(Oligostachyum lubricum)叶片光合色素、可溶性蛋白、脂质过氧化和抗氧化系统的影响研究.结果表明:处理90d后,随着O3浓度的升高,毛竹和四季竹叶片叶绿素(Chl)、类胡萝卜素(Car)含量均呈下降趋势,但毛竹的降幅大于四季竹.MDA含量、POD活性呈升高趋势,毛竹的升幅大于四季竹.SOD活性毛竹呈下降趋势,而四季竹表现出先升高后降低的规律.可溶性蛋白含量毛竹和四季竹分别呈升高、降低趋势;与CF相比,低O3浓度处理(≤50 nl·L-l)并未对毛竹和四季竹造成明显伤害,而高O3浓度处理(≥100 nl·L-1)下毛竹和四季竹叶片光合色素含量和SOD活性极显著降低,MDA含量和POD活性极显著提高,导致毛竹和四季竹叶片光合色素合成受阻,叶片老化加快,膜脂过氧化程度加剧,膜结构和抗氧化系统功能遭到破坏;主成分分析表明,四季竹对O3胁迫的耐受能力明显强于毛竹.  相似文献

Filter feeding in mussels is a secondary adaptation where the gills have become W‐shaped and greatly enlarged, acting as the mussel filter–pump. Water pumping and particle capture in the blue mussel, Mytilus edulis, have been studied over many years. Here, we give a short status of the present understanding of ciliary structure and function of the mussel filter–pump, supplemented with new photo‐microscope and scanning electron microscopy (SEM) pictures of gill preparations. Pumping rate (filtration) and pressure to maintain flow have been extensively studied so the power delivered by the mussel pump to the water flow is known (1.1% of total respiratory power), but the actual cost based on gill respiration is much higher (19%), implying that the cost of maintaining of the large gill pump is considerable and that only relatively little energy can be saved by stopping or reducing the activity of the water‐pumping cilia so that continuous feeding with a ‘minimal scaled’ pump is cheaper than discontinuous feeding with a correspondingly larger pump. According to the present view, the pump proper is the beating lateral cilia (lc) on the gill filaments and particle capture is accomplished by the action of laterofrontal cirri (lfc) transferring particles from the main water current to the frontal gill filament currents driven by frontal cilia (fc). Unexplained aspects include retention efficiency according to particle size and the role of pro‐laterofrontal cilia (p‐lfc) placed between the lfc and fc. The structure of cilia and the mode of ciliary beating have been re‐examined in this study by new high‐resolution light and scanning electron microscopy of isolated gill preparations exposed to serotonin (5‐HT) stimulation which can activate the lc and lfc at low concentrations (10?6 M), but removes the lfc from the interfilament canals at higher concentrations (10?5 M).  相似文献

厚壁毛竹与毛竹叶表皮微形态特征比较研究   总被引:1,自引:0,他引:1  
利用扫描电镜技术,对厚壁毛竹(Phyllostachys edulis‘Pachyloen’)与毛竹(Phyllostachys edulis)营养叶的表皮微形态进行对比研究,结果表明两竹种叶表皮微形结构组成基本相似,均以下表皮结构较为丰富,分布有乳突,微毛,刺毛,硅细胞,气孔器等结构。两竹种叶表皮主要区别在于厚壁毛竹的气孔轴向长度大于毛竹,而气孔密度要低于毛竹,但差异不显著。而两竹种叶的乳突密度相差不大,在不同年龄的竹叶间也无明显变化规律。同一竹种不同竹龄间竹叶表皮气孔密度和气孔轴向长度却有着显著的差异。1年生厚壁毛竹的气孔密度显著高于2年生竹和5年生竹,气孔轴向长度呈现出"V"字形变化规律。毛竹的气孔密度也出现类似的变化规律,但5年生竹叶的气孔密度最大,气孔轴向长度则表现出随秆龄增大而逐渐增长的趋势,这与竹材物质形成积累规律相一致。通过对比研究厚壁毛竹和毛竹叶表皮形态结构特征,两竹种叶表皮微形态特征相似与两竹种系统分类上的地位相近是一致的,同时对厚壁毛竹厚壁性状形成的结构基础进行探讨,为进一步揭示厚壁形成的机理提供了理论依据。  相似文献

The flat oyster Ostrea edulis is native to Europe and populations have been severely depleted by the parasite Bonamia ostreae since the 1980s. Additional genetic markers are required to improve population genetics study and linkage map development for selection for B. ostrea-resistance in this species. Here, we characterized 27 novel microsatellite loci for O. edulis. Number of alleles per locus ranged from 6 to 25 and observed heterozygosity between 0.375 and 1. Null alleles were suggested at a few loci but most loci were in Hardy-Weinberg agreement enabling their reliable use in further population and mapping genetics approaches.  相似文献

1. Specialist herbivores often become less abundant per unit of host tissue as host density increases (resource dilution). They usually become less abundant when non-host species are mixed with their host plants (associational resistance). Most studies of these trends have involved herbaceous host plants and have not examined both trends for the same herbivores. 2. Three hypotheses were tested for the response of insect specialists to host plant density: resource concentration, plant apparency and resource dilution. Two hypotheses were tested for the response of herbivores to non-host plants: associational resistance and plant apparency. 3. From 1992 to 2007, I examined the responses of three monophagous insect herbivores to the densities of their host, Pinus edulis, and of two non-hosts, Pinus ponderosa and Juniperus spp. 4. Herbivore loads increased with host density, though the correlations were weak and often variable between generations. These results were consistent with the resource concentration and plant apparency hypotheses, but not with resource dilution. 5. Herbivore loads decreased as non-host density increased, consistent with the associational resistance hypothesis. This and other studies have shown that associational resistance is important in many types of plant communities. 6. The absence of resource dilution on woodland trees contrasted with studies of herbaceous host plants. Further comparisons of woody and herbaceous host plants are needed to elucidate the reasons for this difference.  相似文献

Protein thiols are targets of oxidative stress. Their modification was analysed in gill extracts of the mussel Mytilus edulis, exposed to menadione. Diagonal gel electrophoresis revealed two clusters of carbonylated proteins involved in interchain disulphide linkages. Immunoblotting identified these as being associated with protein disulphide isomerase (PDI) and actin and this was confirmed by immunoprecipitation. Protein free thiols (-SH) were identified in 2-DE separations by labelling with 5-iodoacetamidofluorescein (IAF). Cysteines involved in disulphide bridges were identified by blocking free -SH with N-ethylmaleimide, reducing disulphides with DTT and IAF labelling. Several protein spots containing free thiols disappeared on exposure to menadione. Conversely, new protein spots containing disulphides appeared in response to menadione which may be protective against oxidative stress. In-gel tryptic digestion followed by LC/MS-MS and database searching identified some of the free thiol targets: PDI; hsp gp96; calreticulin; heavy metal binding protein. Tubulin, PDI, enolase and gelsolin contained new disulphide bridges in response to menadione. Our findings indicate a protein level response to oxidative stress principally involving PDI, chaperone-like and cytoskeletal proteins. Since many environmental pollutants cause oxidative stress, studies on PDI and structural proteins may be particularly relevant to understanding toxicity in this popular sentinel species.  相似文献

The structure and gelling properties of alkali-modified agar from Gracilaria edulis were investigated. 1H and 13C NMR experiments revealed a basic repeating unit of alternating 3-linked 6- O-methyl-β-D-galactopyranose and 4-linked 3,6-anhydro-α-L-galactopyranose. Partial methylation at O-2 of the anhydrogalactose moiety was also revealed. Meanwhile, the O-4 of the methylated galactose residue was detected to exhibit partial sulfation by NMR and FT-IR spectroscopy. The gel strength and syneresis index of the extracted agar were considerably enhanced by the addition of sodium, potassium, and calcium ions. The ion-driven gelation and peculiar sulfate position conferred the agar's similarity to κ-carrageenan. EMMY is an ecophysiological model of the growth and reproduction of a single mussel (Mytilus edulis L.). It contains feedback loops in the uptake and metabolism of food and in the partitioning of carbon and nitrogen to the internal state variables somatic tissue, storage, organic shell matrix and gametes. In this paper EMMY is used to simulate individual mussel growth in a series of mesocosm experiments with different inorganic nutrient loads (N and P). The experiments explore the impact of eutrophication reduction scenarios on mussel growth under defined and controlled conditions.In earlier studies EMMY was calibrated using expert knowledge on growth and reproduction during a period of 5 years. The resulting calibrated model was validated for system inputs and observations of three ecosystems with significantly different food and silt concentrations. EMMY reproduced the mussel growth sufficiently accurate in ecosystems with moderate or high food concentrations. In this study EMMY was adapted in order to cope with low food concentrations, then recalibrated (using the original calibration data and procedure) and applied without further calibration to 3 replicated mesocosm experiments. The EMMY simulations in this study show the ecophysiological response of mussels to different food (phytoplankton and detritus) concentrations. It is concluded that the mussels can adapt to significantly reduced food concentrations, due to inorganic nutrient load reduction, and still maintain growth.  相似文献

 Using 6 years of observational and experimental data, we examined the hypothesis that water and nutrient stress increase the susceptibility of pinyon pine (Pinus edulis) to the stem- and cone-boring moth (Dioryctria albovittella). At two geographic levels, a local scale of 550 km2 and a regional scale of 10,000 km2, moth herbivory was strongly correlated with an edaphic stress gradient. At a local scale, from the cinder soils of Sunset Crater to nearby sandy-loam soils, nine of ten soil macro- and micronutrients, and soil water content were lowest in cinder-dominated soils. Herbivore damage was six times greater on trees growing in the most water and nutrient deficient site at Sunset Crater compared to sites with well-developed soils. Percentage silt-clay content of soil, which was highly positively correlated with soil nutrient and soil moisture at a local scale, accounted for 56% of the variation in herbivory at a regional scale among 22 sites. Within and across sites, increased stem resin flow was positively associated with reduced moth attack. On the basis of moth distribution across a stress gradient, we predicted that pinyons growing in highly stressful environments would show increased resistance to herbivores if supplemented with water and/or nutrients. We conducted a 6-year experiment at a high-stress site where individual trees received water only, fertilizer only, and water + fertilizer. Relative to control trees, stem growth and resin flow increased in all three treatments, but only significantly in the water + fertilizer treatment. Although there was no significant difference in herbivore damage among these three treatments, there was an overall reduction in herbivore damage on all treatment trees combined, compared to control trees. This experiment suggests that release from stress leads to increased resistance to insect attack and is consistent with our observational data. While other studies have predicted that short-term stress will result in herbivore outbreaks, our studies extend this prediction to chronically stressed host populations. Finally, while flush-feeders are not predicted to respond positively to stressed host plants, we found a positive association between herbivore attack and stressed pinyon populations.
